Harry Potter Wizards Collection brings home all eight movies on a ridiculous 31 discs (video)

Now that all of the Harry Potter movies have been released, Warner Bros. has seen fit to slide them together in one truly epic set. Harry Potter Wizard's Collection spans 31 discs including the theatrical version of each movie, extended cuts of the first two flicks, 3D versions of the last two, Ultraviolet digital copies and several bonus discs with ten hours of new to disc bonus content and 5 hours of never before seen extras. Of course, we should also mention the incredibly detailed box it all comes in, seen in the CG video above. Of course, you can't always have everything, and some fans are upset are upset about what this collection doesn't include -- extended versions of the last six movies. Those still interested can preorder the $400 MSRP set (currently selling $349 on Amazon) for delivery September 7th, and get an early preview of one of the special features embedded after the break.

XBMC Eden officially steps out of beta, available for download now

Been snacking on popcorn with the beta build of XBMC 11.0 Eden since it got released last December? If, you'll be pleased know that the full-on release version is now officially available for download. In case you don't recall, this latest build of the media center house many new features, not limited to Addon Rollbacks (in case you hate their new builds), a plethora of speed-improvements, official "in- sync support" for iOS devices, AirPlay functionality and UI tweaks. There's also good news for Ubuntu users, as XBMCUbuntu Final has been officially announced as the successor to XBMC Live. Presbyterian Court Weighs Property Battle in California

A Presbyterian court, in a case with implications for dozens of congregations exiting a denomination divided over homosexuality, convened on Thursday to decide if a California parish may break away with property worth millions of dollars.

Testimony began before the Synod of the Pacific, consisting of ministers and church elders from throughout the West, on a case stemming from a decision by the Community Presbyterian Church of Danville, an affluent San Francisco suburb, to join a more conservative denomination.

A San Francisco-area church governing body agreed in 2010 to allow the Danville congregation to keep real estate that had been appraised at nearly $14 million after its parishioners voted to split from the mainline Presbyterian Church U.S.A.

Critics of the move, however, have argued that the regional governing body lacked authority to approve the property transfer.

"We are here today because we believe the Presbytery of San Francisco has made an unauthorized gift of property," Joan Blackstone, a retired lawyer and church elder who represents opponents of the property transfer, said in opening statements.

Thursday's trial highlights deep divisions within the parent church and its 2 million members, as well as other mainstream Protestant denominations, over the ordination of gay clergy and blessing of same-sex unions.

Although it does not allow ministers to perform same-sex marriages, the Presbyterian Church allows its ministers to bless gay unions.

The synod's trial, held in a hotel banquet room near the San Francisco Airport, will decide whether the presbytery properly used the church's so-called "gracious-dismissal" policy to allow the Danville parish to leave with its buildings and more than 7 acres of land held in trust for the Presbyterian Church U.S.A.

The committee of local ministers "considered, prayed about it and decided the buildings should continue to be used by the Danville congregation as they continue to do God's work," the Rev. Joan Huff, a San Francisco minister who represents the presbytery, told the tribunal. "Grace is not always fair. Grace cannot be monetized. It cannot be bought, earned or won."

"It if could, it would cease to be grace," she said.

Blackstone urged the 10 commissioners hearing the case to consider the impact of their decision on church groups throughout the nation struggling with how to divide property.

OPENED CLERGY TO HOMOSEXUALS

The Presbyterian Church U.S.A. formally opened the ranks of its clergy to homosexuals last spring in a move that triggered a schism, prompting some opposed to gay clergy to form a new denomination, the Evangelical Covenant Order of Presbyterians.

The Presbyterian Church's pension board further alienated some conservative congregants this month by approving benefits for same-gender partners and their children starting next year.

Upset over differences in biblical interpretation of social issues, including questions of gay life and abortion, the Danville congregation began separating from the parent church in 2010.

They want to join yet another offshoot denomination, the Evangelical Presbyterian Church. That church believes "unrepentant homosexual behavior is incompatible with the ordination vows."

"This case is going to establish a precedent of sorts in the minds of people in this denomination," Blackstone said in her opening trial statement. "It's going to be considered by congregations as they request to be dismissed."

The Kentucky-based denomination has endured other splits in its history, including schisms over slavery and the ordination of women. But the Rev. Gradye Parsons, the highest-ranking official in the church general assembly, said some congregations that have left the fold during the past 20 years have rejoined.

"I think it's sad that people are leaving," he said. "I see it more as a separation than a divorce and hope someday we can be reconciled."

The Danville church, founded in 1865, consists of about 1,800 members.

The Rev. Scott Farmer, the church's senior pastor, served on a task force charged with writing the "gracious-dismissal" policy, Blackstone said. She questioned whether Farmer had a conflict of interest when the Danville church was the first to ask to be dismissed under the policy.

Farmer could not be reached for comment.

A website for the Presbyterian Lay Committee, a conservative advocacy group, listed 35 congregations that had started the process of separating from the denomination since July. The First Presbyterian Church in Colorado Springs, with more than 4,000 members, is the largest to take part in the exodus.

The synod is expected to issue a written decision next week.

Pope says communism does not work in Cuba

ABOARD THE PAPAL PLANE -- Pope Benedict said on Friday communism no longer works in Cuba and the Catholic Church was ready to help the island find new ways of moving forward without "trauma."

Speaking on the plane taking him from Rome for a trip to Mexico and Cuba, the pope told reporters: "Today it is evident that Marxist ideology in the way it was conceived no longer corresponds to reality."


Responding to a question about his visit to the island, a communist bastion 90 miles off the coast of the United States for more than 50 years, Benedict added: "New models must be found with patience and in a constructive way...we want to help."

Benedict, who is due to arrive in Cuba on Monday after a three-day visit to Mexico, called for freedom of conscience and freedom of religion on the island.

Also on Friday, Benedict promised to "unmask the evil" of drug trafficking in Mexico, which has been wracked by a surge in gang violence over the past five years.

'We are all fed up'
The pope gave a forceful response when asked on board his plane about drug trafficking in Mexico, which has generated violence claiming some 50,000 lives since 2007.

"We must do whatever is possible to combat this destructive evil against humanity and our youth," he told reporters. "It is the responsibility of the Church to educate consciences, to teach moral responsibility and to unmask the evil, to unmask this idolatry of money which enslaves man, to unmask the false promises, the lies, the fraud that is behind drugs."

The relentless bloodletting was in the mind of many of those waiting to see Benedict in Leon, a Roman Catholic stronghold that has avoided the worst of the brutal turf wars between drug cartels and clashes with security forces.

"The Church has to address the violence, give us a message that there can be change. We are all fed up, our society has been hurt," said Ruben Santibanez, a local doctor.

Christophe Pierre, the Vatican's diplomatic representative in Mexico, said Benedict would not ignore Mexico's plight.

"Violence is being talked about, we can't hide it, but there is a lot more to Mexico than the violence, and the desire to live in harmony is precisely what the Holy Father is coming for," he said on Thursday.

Crowds of devout Mexicans readied campsites in Leon in preparation for a massive outdoor mass on Sunday where hundreds of thousands of people are expected.

Benedict hopes to rally the faithful in the world's second most populous Catholic nation as more Mexicans are lured to evangelical Protestant churches.

But the German pope faces a challenge generating the same kind of fervor as his charismatic Polish predecessor, Pope John Paul, who was beloved in Latin America and drew pulsating crowds whenever he criss-crossed the region during his 27-year papacy.

Abuse scandals
Highlighting the growing religious divisions in Mexico, a large Protestant church in Leon held a service with hundreds of worshipers on Thursday, prominently displaying a sign that read "We are not Roman Catholics."

The Vatican has been stung by accusations of child abuse by priests in parishes around the world - including revelations about Mexican religious leader Marcial Maciel, who founded a prominent Catholic order, the Legionaries of Christ.

Maciel, who died in 2008 at the age of 87, was disgraced by allegations of sexual abuse and drug addiction.

During the pope's visit, religious scholar Bernardo Barranco will present a new book by ex-Legionaries with scores of leaked Vatican documents the authors say prove the Holy See knew about Maciel's molestation of young boys and morphine use for decades.

The hacker group Anonymous in Mexico crashed at least two of the websites for Benedict's visit to Mexico on Thursday, claiming his trip is a political move to support the president's party.

Anonymous Mexico said in a video posted on social media sites that the pope's visit will cost Mexicans money that could be better spent on the poor, and is meant to support the PAN in the July 1 presidential election. PAN candidate Josefina Vazquez Mota trails front-runner Enrique Pena Nieto of the Institutional Revolutionary Party by at least 10 percentage points in most polls.

Tehran forces Iranian Jews to join anti-Israel Global March

The Islamic regime in Tehran was not satisfied with the public support the Iranian Jewish community?s was forced to confer on the Global March to Jerusalem for which Iran is recruiting Islamists worldwide. The delegations are scheduled to mass on the Lebanese and Jordanian borders with Israel and at West Bank and Gaza checkpoints on March 30, when Israeli Arabs mark Earth Day every year.

Iran?s ancient Jewish community of around 15,000 souls (9,000 in Tehran, 4,000 in Shiraz and 1,300 in Isfahan) has been living in fear of reprisals should Israel or the United States carry out a military operation against the country?s nuclear facilities. ?Now, they face a fresh danger of murder and abduction by Hizballah and Palestinian gunmen and terrorists in Lebanon.
Jewish communal leaders were instructed by the Iranian authorities this week to have at least 10 young men aged 18 to 22 ready for the march. Prostate cancer is the most common malignancy in men .Both the morbidityand mortality rate from prostate cancer rise almost exponentially in recent years.Many researches showed that its tumorgenesis and biological characters are closelyrelated to many genes. Growing evidence suggests that many kallikreins areimplicated in it. Human kallikrein gene 7(KLK7; HSCCE), also named HSCCE/PRSS6, is a new member of the human kallikrein gene family, which was found inskin first, had drawn great attention and been undertaken wide studies in manymalignant tumors.Objective The aim of this study were : (1) to determine whether KLK7 gene isexpressed in prostate cell line PC-3. (2) to investigate whether DHT had theeffects on KLK7 mRNA level in PC-3 cell in vitroMethods (1) Total RNA was extracted from human prostate cell line PC-3 andhuman kallirein7(KLK7) gene cDNA was amplified by PCR after reverse-transcription using Oligo (dT) primer. Amplified products was ligated to PGEM-TEasy plasmid vector. The recombinant plasmid was transfered to the competentE.coli DH5?. Indentified by enzyme digestion, KLK7 gene in recombinant plasmidwas sequenced by sanger dideoxy-mediated chain-termination method. (2)Thehuman prostate cell line PC-3 were cultured at 37?in a CO2 incubator with F12culture medium and were divided into seven groups by different DHT stimulatingconcentrations of 0M?10-9M?10-8M?10-7M?10-6M?10-5M and 0.01% Ethanol.Following culture in day 1, day 2, day 3 and day 4, MTT method was applied toobserve the cell proliferation . RT-PCR was engaged to find the KLK7 level in thePC-3 cell under the different concentrations of DHT stimulation.Results (1)The cloned kallikrein7 gene is about 762 bp in length. A sequencecomparison of this with the reported kallikrein7 sequence in the Genebankindicated that the cloned prostate cell line PC-3 KLK7 gene has two nucleotidedifference. (2) In day 1 of culture, the cell treated with 10-8mol/l DHT grew fast,there was significant increase of OD in this group compared with the control group( P < 0.05). In day 4 of culture , there were significant increase of OD in thegroups treated with 10-9mol/l?10-8mol/l?10-7mol/l?10-6mol/l DHT compared withthe control group ( P < 0.05) except the group treated with 10-5mol/l DHT (P >0.05). (3) RT-PCR showed that the group treated with 10-9mol/L ?10-8mol/L?10-7mol/L?10-6mol/L DHT had no KLK7 mRNA expression. TheKLK7 mRNA OD value in control group and 10-5mol/L DHT group was 0.9061?0.9367 respectively compared with control gene .Conclusion Our study indicated: (1) KLK7 gene is expressed in prostate cellline. It may be provide the evidences for further studies of the KLK7 gene inprostate cancer. (2) Lower concentration of DHT can enhance the proliferation ofPC-3 cell. Its optimal concentration is 10-8mol/L. (3) the expression of KLK7mRNA may be regulated by androgens and has certain relations with prostatecancer cell growth. Further studies are required to prove this hypothesis.
